Asymbiotic Germination of Cymbidium ensifolium


Abstract
The study was conducted to determine the optimal basal media and the concentration of plant growth regulator for germination of seeds and shout emergence from rhizomes of oriental orchid, Cymbidium ensifolium.
1. Germination u-as similar in light and dark condition, but the growth of rhizome after germination was better under dark than under light condition in several media. Germination was best in Tsuchiya & Nitsch, Kyoto solution II and Nf-G medium among the 14 media tested. In Nitsch. Tsuchiya and Kyoto solution I, germination was good only under dark condition. Growth n rhizome after germination was also best in the three best media for germination. In Kyoto solution I , it was better under dark than light condition.
2. Proliferation of rhizome in the media for differentiation eras good in Nf-G medium containing 2.0ppm NAA and 0.2ppm or 2.0ppm kinetin, that time the. rhizome used was subcultured in Tsuchiya & Nitsch¢¥s medium. Both proliferation of rhizome and growth of shoots differentiated from rhizome, the rhizome used was subcultured in Nf-G medium, were best in Nf-G medium containing 2.0ppm NAA and 0.2ppm kinetin.
3. The number of shoots, rhizome was most in the Nf-G medium containing 2.0ppm NAA and 0.2ppm kinetin.
